What Peptides Actually Do for Skin
"Peptides" is a broad umbrella term. The honest answer to what they do depends entirely on which peptide you're talking about. Skincare marketing tends to treat all peptides as interchangeable evidence for "anti-aging." But the four main functional categories work through genuinely different mechanisms, with very different amounts of research behind them.
Signal peptides, Matrixyl, collagen production & elastin
This is the largest and best-studied group. It includes ingredients like palmitoyl pentapeptide-4 (often branded Matrixyl or Matrixyl 3000) and palmitoyl tripeptide-1. The theory is that these short peptide fragments mimic pieces of broken-down collagen. Skin cells respond to their presence by ramping up collagen production and elastin, essentially a feedback signal that damage has occurred and repair is needed. Lab and some small clinical studies show measurable increases in collagen markers with consistent use over 8-12 weeks. Most published research on specific signal peptides comes from the ingredient manufacturers themselves, rather than independent, large-scale trials. Formulas like The Ordinary's Buffet and Medik8's Liquid Peptides blend several signal peptide complexes in one serum.
Carrier peptides: copper peptides, wound healing & skin barrier support
Copper peptides (glycyl-histidyl-lysine bound to copper, often listed as "copper tripeptide-1") originated from wound healing research, where much higher concentrations were shown to support tissue repair in clinical settings. Skincare formulations use far lower concentrations, so the wound-healing data doesn't translate directly. There's a reasonable case that copper peptides support the skin barrier and may have some antioxidant benefit at cosmetic doses. Naturium and Paula's Choice both formulate multi-peptide serums that pair copper peptides with niacinamide for extra barrier support. This is one of the more debated peptide categories, partly because copper can also act as a pro-oxidant in the wrong context. That's why formulators tend to use it carefully, in combination with other stabilizing ingredients.
Neurotransmitter-inhibiting peptides, Argireline & the "Botox alternative" claim
Acetyl hexapeptide-8 (Argireline) is the ingredient behind most "instant Botox in a bottle" marketing claims. The mechanism is real in concept. It's designed to interfere with the release of a neurotransmitter involved in muscle contraction, the same general pathway botulinum toxin injections target, just far more mildly and topically instead of injected. The problem is scale. Injectables work directly on the neuromuscular junction. A topical peptide has to survive being applied to the skin surface and somehow reach a comparable depth in a meaningful concentration, which is a much harder ask. The clinical trials that exist tend to be small, short, and often industry-funded. They show modest reductions in wrinkle depth at best, nowhere near injectable results. It's a legitimate ingredient to try for subtle softening around expression lines, like the forehead or crow's feet. But the "alternative to Botox" framing oversells what the data supports.
Enzyme-inhibitor peptides, MMPs & clinical studies on wrinkle depth
Rather than stimulating new collagen, this category aims to slow the enzymatic breakdown of collagen that already exists. It includes ingredients targeting matrix metalloproteinases (MMPs). It's a less commonly marketed category, and evidence is even thinner than for signal peptides. But the mechanism, protecting what's there rather than adding more, is a reasonable complementary approach in a formula that also includes signal peptides.
What this means for expression lines & realistic skincare expectations
Across all four categories, a few honest patterns hold. First, molecule size and formulation matter enormously. A peptide has to be stabilized and delivered in a way that lets some of it penetrate the skin's outer layer. Not every product on the market does this well, or discloses how. Second, results, where they exist, take weeks to months of consistent use to become visible, not days. Third, most of the strongest peptide research is short-term, small-scale, or funded by the companies that sell the raw ingredient. That's a real limitation compared to the far larger body of independent research behind retinoids. None of that means peptides don't do anything for expression lines and firmness. It means "supports" and "may help" are more honest words in skincare than "transforms" or "replaces."
